The EMA's human-medicines committee has recommended EU marketing authorization for Ascendis Pharma's once-weekly navepegritide, to be sold as Yuviwel, for children aged 2 and older with achondroplasia. The CHMP adopted the positive opinion at its 12–15 October 2026 meeting and Ascendis confirmed it on 16 October; a final European Commission decision is expected before the end of the year, eight months after the same drug won accelerated approval in the United States.

A second yes, five months after the first

Ascendis Pharma submitted its EU Marketing Authorisation Application for navepegritide in 2025, after the drug cleared the FDA's accelerated-approval bar on 27 February 2026 in the United States. The CHMP's 16 October 2026 opinion covers the same population FDA approved: children two years and older with achondroplasia whose growth plates remain open. A final decision from the European Commission, which normally follows CHMP's recommendation within roughly two months, is expected before the end of 2026.

The approval basis in both regions rests on the same evidence: three randomized, placebo-controlled trials, including the pivotal ApproaCH study, plus up to three years of open-label extension data. At week 52 of ApproaCH, children on navepegritide reached an annualized growth velocity of 5.89 cm against 4.41 cm on placebo — the superiority finding both FDA and CHMP relied on.

“We are pleased to receive this positive opinion reflecting the unique profile of TransCon CNP.” Jan Mikkelsen, President & CEO, Ascendis Pharma — 16 October 2026 announcement

Why a prodrug, not another daily shot

Achondroplasia is caused by a mutation that leaves the FGFR3 growth-plate receptor permanently switched on, slowing bone growth. C-type natriuretic peptide (CNP) counteracts that signal, but natural CNP breaks down in the body within minutes. BioMarin's Voxzogo (vosoritide) is a CNP analog dosed once daily to work around that half-life. Ascendis built navepegritide as a TransCon — its own prodrug technology — designed to release CNP continuously from a single weekly injection rather than a daily peak and trough.

Ascendis has not published a trial comparing navepegritide directly against Voxzogo, so the once-weekly dosing claim rests on the two drugs' separate pharmacokinetic profiles rather than a head-to-head growth result.

Three things that set navepegritide's pitch apart

One injection a week

TransCon's prodrug chemistry is designed to release CNP continuously, aiming to replace Voxzogo's daily injection with a weekly one — though Ascendis has not run a head-to-head growth trial to prove the clinical difference that makes.

Beat placebo, not the incumbent

The 5.89 cm vs 4.41 cm annualized growth velocity result in ApproaCH is what both FDA and CHMP approved on. It establishes navepegritide works; it does not establish it works better than Voxzogo.

A platform, not a one-drug company

Navepegritide is Ascendis's third TransCon product to reach the market, after SKYTROFA (growth hormone deficiency) and YORVIPATH (hypoparathyroidism), both already generating revenue.

BioMarin's eight-year head start

Achondroplasia has had exactly one approved drug since 2021: BioMarin's Voxzogo, now sold in 55 countries and guided to $900–935 million in revenue for 2025, up 24% year over year through the first nine months. Navepegritide's US approval in February 2026 ended that monopoly; a positive CHMP opinion extends the challenge into BioMarin's largest market outside the US, where roughly three-quarters of Voxzogo's revenue is booked.

Regulatory News outlook

We expect the branded achondroplasia treatment market to reach roughly $1.6 billion a year by 2030, with navepegritide taking a meaningful minority of new patients alongside Voxzogo rather than displacing it.

How we got here: The midpoint of BioMarin's $900–935 million 2025 Voxzogo guidance ($917 million), compounded at 12% a year — about half its reported 24% year-over-year growth rate — for five years to 2030, reaches roughly $1.6 billion from the incumbent alone, before any navepegritide sales are added. That is above The Business Research Company's $0.98 billion 2030 estimate, which we treat as a floor, and within reach of Strategic Market Research's higher $2.4 billion figure.

What Q4 still has to settle

A CHMP positive opinion is a recommendation, not an authorization: the European Commission makes the binding decision, typically within about two months of CHMP's vote, and it has not historically departed from a CHMP recommendation for a product with this level of trial support. Ascendis's own US approval was also an accelerated one, meaning continued US marketing could depend on confirming the growth-velocity benefit translates into a longer-term clinical outcome in further study — a condition that sits alongside, not instead of, the EU process now underway.

  • EC decision: expected in the fourth quarter of 2026, following the CHMP's positive opinion.
  • First EU launch: Germany, targeted for the first quarter of 2027, contingent on EC authorization.
  • US accelerated approval: continued approval may depend on confirmatory evidence of clinical benefit beyond annualized growth velocity, per the FDA's own accelerated-approval framework.
  • Vision 2030: Ascendis has set a company-wide target of €5 billion in revenue by 2030, built on SKYTROFA, YORVIPATH and navepegritide together — not a navepegritide-specific figure.

Frequently asked questions

What did the CHMP recommend?

On 16 October 2026 the EMA's Committee for Medicinal Products for Human Use issued a positive opinion recommending EU marketing authorization for once-weekly navepegritide (to be sold as Yuviwel) for children aged 2 and older with achondroplasia whose growth plates are still open. A final European Commission decision, which normally follows CHMP's recommendation, is expected in the fourth quarter of 2026.

Is Yuviwel already approved anywhere?

Yes. The FDA granted it accelerated approval in the United States on 27 February 2026 for the same indication, and Ascendis said the drug became commercially available there in April 2026.

How is navepegritide different from BioMarin's Voxzogo?

Both are C-type natriuretic peptide (CNP) therapies for achondroplasia, but Voxzogo (vosoritide) is dosed once daily while navepegritide is engineered as a prodrug intended to provide more continuous CNP exposure with a once-weekly injection. Ascendis has not published a head-to-head trial against Voxzogo.

When could EU patients get the drug?

Ascendis has said it plans a first EU launch in Germany in the first quarter of 2027, contingent on the European Commission's final decision, expected in the fourth quarter of 2026.
